Human DNA Polymerase Alpha Pyrophosphate Assay age
A colorimetric assay
The colorimetric assay for human DNA polymerase alpha is based on detection of the pyrophosphate molecules generated by the DNA polymerase reaction. The pyrophosphate product is converted to phosphate by pyrophosphatase. The final product phosphate is measured by light absorbance at 650 nm (A650) using the phosphate detection reagent. The assay is in a 384-well or 96-well plate format designed for high throughput screening of compound collections. It is suitable for tests of human DNA polymerase alpha and its inhibitors.

The Human DNA Polymerase Alpha Pyrophosphate Assay Kit (Catalog No. HDPA100KP) includes all the assay kit components except the enzyme for 100 assays in a 384-well plate assay format: 400 ul of 10 x Buffer, 30 ul of 100 x DNA, 30 ul of 100 x dNTP, 30 ul of 100 x pyrophosphatase (PPase) and 6 ml of Dye.
