Bacteriophage DNA ligases repair single stranded nicks in duplex DNA and some DNA/RNA hybrids. The ligases catalyze the formation of a phosphodiester bond between 5' phosphate and 3' hydroxyl termini in duplex DNA or RNA. They are frequently used in DNA ligation in molecular biology.  The DNA ligase assay is to measure the DNA ligase product in which the diphosphodiester bond is formed at the single stand break of a duplex DNA substrate.  The ligase reaction is monitored by the fluorescence intensity at 535 nm.  The assay is in 96-well plate format and can be used for measurement DNA ligase activity and testing their inhibitors.

The Bacteriophage T4 DNA Ligase Assay Kit (Catalog number T4L100K) includes 500 µl of 10 x buffer, 44 µl of 100 x DNA substrate, 44 µl of 100 x ATP (2 mM), 21 ml of Reagent T and 220 µl of 10 x dye for 100 assays.  The kit does not include ligase.

The Bacteriophage T4 DNA Ligase Assay Kit Plus (Catalog number T4L100KE) includes 500 µl of 10 x buffer, 44 µl of 100 x DNA substrate, 44 µl of 100 x ATP (2 mM), 21 ml of Reagent T, 220 µl of 10 x dye  and 44µl of 100 x T4 DNA ligase (5000 U/ml) for 100 assays. 

The Bacteriophage T7 DNA Ligase Assay Kit (Catalog number T7L100K) includes 500 µl of 10 x buffer, 44 µl of 100 x DNA substrate, 44 µl of 100 x ATP (2 mM), 21 ml of Reagent T and 220 µl of 10 x dye for 100 assays.  The kit does not include ligase.
